Tango Therapeutics used ESMO to commit to a registrational programme in one of the hardest cancers to treat. In the Phase 1/2 study of vopimetostat, its oral PRMT5 inhibitor, combined with Revolution Medicines RAS inhibitor daraxonrasib, 18 of 22 evaluable patients with MTAP deleted, RAS mutant metastatic pancreatic cancer treated in the second or third line responded, an objective response rate of 82 percent. The disease control rate was 100 percent, median progression free survival had not been reached, and 88 percent of patients were alive and free of progression at nine months, at an August 20 data cutoff.
The comparison is what drew analyst attention. Revolution own data for its RAS inhibitor alone showed response rates of 29 percent in second line and 20 percent in later line pancreatic patients in the reporting, which is why analysts described the combination as highly synergistic. A second combination, vopimetostat plus the G12D selective zoldonrasib, produced a 54 percent response rate in 35 evaluable patients, with a 97 percent disease control rate.
The shares still fell 16.4 percent to 23.34 dollars. The 82 percent rate was lower than the 92 percent reported in June in a smaller group of 12 patients, and seven of the 18 responses were unconfirmed at the cutoff, though all were confirmed afterwards. Alongside the data, Tango disclosed in an SEC filing that it will cut about 14 percent of its workforce, mostly in research, deprioritise novel target discovery and stop some preclinical work to focus resources on vopimetostat and its follow on candidate TNG456.
The execution risk is explicit. Tango plans a global front line Phase 3 trial of the combination against standard chemotherapy starting in the first half of 2027, but chief executive Malte Peters said the company does not yet have a supply agreement with Revolution for that trial, and that its plan includes continuing work with Revolution and other options. The data support the trial. Whether Tango can run it on a partner drug it does not control is the question the selloff was pricing.
